i think i have to scale for that size

that's a big banner ;)
Ask the printer what dpi is needed at final output then scale down accordingly. aka output will probably be in the 32-75dpi range at most for raster unless the printer accepts the files broken into strips.
Stay vector as much as possible, the scaling then is pretty much guranteed to turn out.

They will end up thanking you for taking the time to talk to them to get the right process files to them and in what scale.

Something that big probably won't be over 35dpi. It might rather be in ipd. LOL.
Definitely talk to the printer. Vendors who print that large have spec sheets giving you scale and resolution instructions that you must follow to get it right.

Are you just randomly posting questions to learn?
Right now the only people to purchase 45' x 300' banner would be to the top corporations in the world and they would only go to an established large format printer who already knows how to put these together. And would probably have top designers to set up the files to print.
You are asking a lot of print questions that many of us say to ask a printer. But are these projects real life projects? Or just ideas that you want to learn how to put together?
We are a very helpful bunch and are willing to answer most questions. But you are not going to get the education on the print world by asking us. You'll need to ferret out more information than we can give you on here. I may be way off, but this seems the trend for your posts.
If you want information on how to get started in the printing and graphic design world, we can direct you to proper information on the subjects. But we need more information than what you are providing through these questions.

Drazan, not only top corporations do things that big. I've worked on a few museum exhibit or arena events that wanted something about that size. And the clients go to a design firm for the design. One can only hope the design firm calls the printer or exhibit builder before finalizing the illustrations and digital files.

You have 300 feet wide billboards in your neck of the woods, Rick? Wow. The ones out here are usually 48 feet by 14 feet, or less.
