I am pretty new to indesign and have worked out your general page set up stuff.
I've been asked to design a brochure that unfolds to a A2 size poster. I'm very unsure about how to set this up. Do I design it all on an A2 with guides to divide it up or do I have 4 A4s for the actual brochure part and an A2 for the poster on the 'back'? If it is the second option - should these be in separte files?
Any assistance with this problem would be greatly appreciated. Thanks.
One single A2 sheet, for both sides - with guides to divide it up.

I generally contact the printer first with any project set up I am unsure of. They can tell you how they like their files set up.

How to design A2 Poster Brochure? Well most that I have seen are an A2 that is quarter folded. Then the brochure may be a half or trifold of that. First I would ask whether they want a tri-fold brochure, 6 panel, or half fold, 4 panel (most tri fold).
I would recommend taking a sheet, folding it down like that. Mark all your panels into a mock-up by justr labeling or sketching them. Notice the brochure will have an inside, that's the poster. If you mock it up, what I'm saying will become clear. Be sure you allow for creep on your panels, especially since it will be folded many times, your inside panel will have to be shorter than the front and back panels, follow me? Also, tou will need to have the piece UV or aqueous coated so that the folds won't crack, making the poster folds show up after it's on the wall. HTH

use the piece of paper solution mkentioned as you will find thaat some of your quarters will be upside down. Folding a piece of paper will hel p you see how you need to make the A2 panels
